1. Adsorption: Pass the juice extracted from the pineapple stem through the adsorption column for adsorption, controlling the flow rate at 800-1000 L/h.

2. Elution: Stop loading the column when the collected volume reaches 5 billion enzyme activity units. After stopping loading, neutralize the adsorbent with 15% NaOH until the effluent pH reaches 7.0. Drain the remaining liquid, then soak in a 7-9% NaCl solution for 30 minutes for elution. Control the elution flow rate and collect the eluate.

3. Acidification and Salting-Out: Add hydrochloric acid to the eluate to adjust the pH to 5.0. Then, add sulfate (usually ammonium sulfate) at 25% by weight of the filtrate. Once completely dissolved, allow to stand at 4°C for 12 hours.

4. Centrifugation and Washing: Centrifuge the mixture after salting-out, and rinse the precipitate 2-3 times with 4°C purified water to obtain crude bromelain.

5. Dissolution and purification: Place the crude enzyme in a dissolution tank and add 10 times the amount of purified water. While stirring, adjust the pH to 7.0-7.5 with 20% NaOH to completely dissolve the enzyme. Once the solution becomes clear, filter to obtain the filtrate.

6. Refining and precipitation: While stirring, slowly adjust the pH of the filtrate to 4.0 with hydrochloric acid. Let it stand for 1 hour to allow the enzyme to precipitate again.

7. Centrifugation: Centrifuge the precipitated enzyme mixture from the refining tank for 20-30 minutes. Rinse the precipitate 2-3 times with cold purified water to obtain the wet bromelain product.

8. Sieving and vacuum drying: Pass the wet product through a 20-mesh sieve, then vacuum dry it at room temperature for 4-8 hours under a vacuum of -0.08-0.09 MPa, controlling the moisture content to below 0.2%, to obtain the dry product.

9. Screening and packaging: The dry product is screened through a 100-mesh sieve, packaged at 20kg/barrel or 25kg/barrel, tested, and put into storage.
